Residual host cell proteins (HCPs), including lipases, can contribute to polysorbate degradation, creating challenges for therapeutic antibody formulation stability.
In this conference presentation, Demeetra scientist Thomas Hart shares research on the development of the CleanCut GS Low HCP cell-line series, using Cas-CLOVER gene-editing technology to simultaneously knock out two lipase genes, LPL and LIPA, in CHO cells.
Hart presents experimental results demonstrating high gene-editing efficiency, no detectable lipase activity in tested engineered clones, and up to 80.6% protection against polysorbate degradation in a 14-day assay compared with parental cells.
Watch the presentation to explore the findings, their potential implications for biopharmaceutical manufacturing, and the next steps in developing CHO cell lines with reduced HCP-related risks.
