The new Harbor-IN format helps biopharma developers establish production cell lines using animal-origin-free transposase mRNA from the start, while maintaining the platform’s stable, nonviral integration and license-free commercial access.
Demeetra has introduced Harbor-IN Elite, an animal-origin-free transposase mRNA format designed for commercial cell line development programs progressing toward GMP manufacturing.
Harbor-IN Elite provides Harbor-IN’s nonviral stable integration system in a non-animal, non-human-origin mRNA format. This allows biopharma developers to address material-origin considerations when the production cell line is first established, rather than revisiting those decisions later as a program advances toward manufacturing.
Material origin matters from the beginning
Cell line development is more than an early research step. For many biologics programs, the cell line established during development becomes the foundation for subsequent process development, characterization, cell banking, and manufacturing.
That makes early material choices important.
Harbor-IN Elite is manufactured using synthetic or plant-derived materials, with no animal- or human-origin materials used in the manufacturing process. By addressing material origin earlier, at the cell line development stage, developers can create a clearer path to later-stage manufacturing requirements.
“The decisions made when a production cell line is first created follow a program through development and manufacturing,” said Christian Cummings, Demeetra Global Business Development Manager. “Harbor-IN Elite gives developers the option to use animal-origin-free transposase from the beginning, while continuing to use the Harbor-IN integration platform as their programs advance.”
Harbor-IN performance and commercial flexibility
Harbor-IN is proven for stable gene integration in biologics production, favoring integration into transcriptionally active genomic regions to support high and durable expression.
The platform accommodates genetic cargo above 200 kb, supporting applications ranging from conventional monoclonal antibodies to more complex multigene constructs such as bispecific antibodies and fusion proteins. Because the transposase is delivered as mRNA, its expression is transient while the integrated expression cassette remains stable. This provides a nonviral approach to creating stable, high-expressing cell populations for further evaluation and clone selection.
Harbor-IN is also available for unrestricted commercial use without a platform license, milestone fees, or royalties, giving developers a straightforward path from cell line development toward commercialization.
Evaluate production potential earlier
Harbor-IN-generated stable pools can be evaluated for productivity before single-cell cloning, providing developers with an earlier view of production potential.
Selected cells can then progress through single-cell cloning, characterization, stability assessment, process development, and cell banking as the program advances toward GMP manufacturing.
Harbor-IN Elite adds animal-origin-free transposase mRNA to that path, allowing teams to address both integration strategy and material origin early.
Built for commercial cell line development
Harbor-IN Elite can support stable integration for a range of biologics applications, including:
- Monoclonal antibodies
- Bispecific and multispecific antibodies
- Recombinant therapeutic proteins
- Multigene expression systems
Supporting documentation for Harbor-IN Elite includes material-origin information to help development teams understand the materials used in manufacture.
